چکیده

It was found that rat kidney contains a protein similar to that previously described in rat liver which catalyzes the transfer of the methyl group from O6-methylguanine in DNA to a proteinbound cysteine residue. The amount of the renal O6-methylguanine-DNA methyltransferase was increased up to 2.5-fold during renal hypertrophy in response to unilateral nephrectomy or treatment with folie acid. These results indicate that the protein in kidney resembles that in rat liver which is known to be increased in response to a variety of hepatotoxins or to partial hepatectomy. The liver O6-methylguanine-DNA methyl transferase was reduced by hypophysectomy or thyroidectomy and could be increased by treatment with growth hormone or thyroxine. The level in the liver was considerably lower than the adult value in 1-day-old rats and increased to adult values by 14 to 21 days. At no time was the amount in the neonatal rat liver higher than in the adult, indicating that liver cell proliferation alone is not obligatorily coupled with an elevated methyltransferase level. The high sensitivity of neonatal rats to liver carcinogenesis by dimethylnitrosamine may be related to the high rate of cell proliferation and the lower capacity to repair O6-methylguanine.

O6-Methylguanine-DNA Methyltransferase and ATP-Binding Cassette Membrane Transporter G2 Promotor Methylation: Can Predict the Response to Chemotherapy in Advanced Breast Cancer?

Background: ATP-binding cassette membrane transporter G2 (ABCG2) gene is one of transporter family and well characterized for their association with chemoresistance. Promoter methylation is a mechanism for regulation of gene expression. O6-Methyl guanine DNA methyl transferase (MGMT) gene plays a fundamental role in DNA repair. MGMT has the ability to remove alkyl adducts from DNA at the O6 pos...

Stability and capacity of dimethylnitrosamine-induced O6-methylguanine repair system in rat liver.

Repeated administration of dimethylnitrosamine (DMN) (2 mg/kg for 3 weeks) to BD IV rats results in an increased capacity for the liver to repair O6-methylguanine in DNA, whereas other DNA adducts (7-methylguanine and 3-methyladenine) are not affected. In the experiments reported here, data on the rapidity of action of the enhanced system, its capacity after different challenging doses of [14C]...

بررسی نقش پلی مورفیسم‌های ژن O6 – متیل گوانین متیل ترانسفراز در سرطان کولورکتال

Abstract Background: Sporadic colorectal cancer is the fourth most common cancer in Iran. The DNA repair protein O6-methylguanine-DNA methyltransferase (MGMT) is involved in the cellular defense against alkylating agents. Genetic alterations in the MGMT gene may impair the protein’s ability to remove alkyl groups from the O6-position of guanine, thereby raising the mutation rate and increasi...

Irradiation-induced expression of O6-methylguanine-DNA methyltransferase in mammalian cells.

O6-Methylguanine-DNA methyltransferase (MGMT) is a DNA repair protein which plays an important role in chemotherapy, mutagenesis, and carcinogenesis. The specific activity of MGMT in female rat liver can be induced by approximately 20-fold by treatment of the rats with gamma-irradiation. Maximum response occurred 48 h after 15 Gy irradiation. MGMT levels in male rats were induced by only 3-fold...
